Objectives: To evaluate the role of caveolae transcytosis ; in large solute transport across capillary endothelium, we assessed the clearances of 125 I-albumin HSA ; mol.radius; ae 36 ; , 131I-IgM ae 120 ; , and polydisperse high MW FITC-Ficoll-400 ae 20-100 ; , across peritoneal capillaries in vivo in Caveolin-1 Cav-1 ; deficient mice. Methods: Male Cav-1 ; KO ; mice were anesthetized and 2 % Isofluorane ; one vein and one artery were cannulated for sampling and infusion purposes. A peritoneal dialysis model was employed, using 2.5 ml of dialysis fluid in the peritoneal cavity for 2 hrs. 125I HSA and 131 I-IgM, and during the last 40 min, FITC-Ficoll-400 were given i.v. Sampling of plasma and dialysate was performed for determination of radioactivity, glucose and FITC-Ficoll concentrations. Conclusions: In Cav-1 ; mice, relative to wild-type mice, the transcapillary Cl of macromolecules was 40 % increased, indicating a higher macromolecule flux through large pores in the KO mice, while the pore radii were unchanged. Data indicate that the transport of albumin, IgM and high MW Ficoll, are independent of the presence of caveolae, but can be predicted to occur by filtration through functional large pores or radius ~200.
